由傳統中藥材黃芩根部所萃取的類黃酮化合物-黃芩素(Baicalein),在許多文獻中已證實具抗氧化、抗發炎與抗癌的活性。在本研究中,我們探討黃芩素對於乳癌細胞的化學預防效果。使用紅油O(Oil Red O)及尼羅紅(Nile red) 螢光染色,觀察到黃芩素誘發乳癌細胞內油滴以及中性脂肪的產生隨著時間與濃度的增加而增加。利用西方墨點法偵測乳房上皮細胞分化指標蛋白的變化,發現黃芩素可誘發乳癌細胞內Milk fat globulin和Cytokeratin 8 等分化指標分子表現量增加。進一步發現黃芩素所誘發乳癌細胞分化的現象受HER2過表現而降低。綜和實驗結果證實黃芩素作用於乳癌細胞株,具引發乳癌細胞分化的活性。
英文摘要
Baicalein, a flavonoid contained in the herbal medicine, extracted from root of Scutellaria baicalensis was found to exert antioxidant, anti-inflammation and antitumor activities. Here, we explored the chemopreventive activity of baicalein in breast cancer cells. By using Oil Red O and Nile red staining, we found that baicalein caused neutral lipids accumulation in dose- and time-dependent manners. Besides, induction of milk fat globulin and cytokeratin 8, makers of mature breast cells, after treating with baicalein were also observed. Furthermore, introduction of HER2 into these breast cells reversed the baicalein induced differentiation. Taken together, our data indicate that baicalein suppresses growth and induces differentiation of breast cancer cells.
